I got the 4th one by beating the game, but I don't know how to get the 5th ship. Is it unlocked by beating the game on 2nd Loop?

That is probably the worst use of the spoiler tag I've ever seen. If you are going to cover how to unlock the 4th ship, you should have covered "beating the game again".

At any rate, yes. Just beat the game again.

On 2nd Loop on Stage 2. Man, 2nd Ship/Wide-Attack helps SO much. The game is definitely harder the 2nd time around, even on normal.

You don't have to beat it twice in a row. You just have to beat the game twice.

I love the fact that loop 2 is significantly different in terms of the level layout.

You can beat it in loop 1 again to unlock E.
