I can't seem to figure out just how many Pokemon are in this game. Not new ones. All of them.
The Sinnoh Pokedex contains exactly 151 Pokemon. There are also 3 extra Event Pokemon you can gain from Nintendo events. Meaning the total of Pokemon you can gain strictly from Diamond and Pearl is 154.
Once you get the National Pokedex and can trade with your old GBA games (Ruby, Sapphire, Emerald, FireRed and LeafGreen), you can port all the old Pokemon over making it a grand total of 493 Pokemon available.

Not only can you port them over, but once you have your Nationaldex you can catch pokemon from specific games by inserting the cart into your DS and finding them in the wild.
Staraptor @ Choice Band/Life Orb
Jolly nature
252 ATK EV, 252 SPD EV, 6 HP EV
Intimidation
- Brave Bird
- Close Combat
- Double-Edge/Return
- U-Turn (put him up-front for a damage+switch-in for something that counters the enemy poke)

Oh yeah, I forgot about that feature.
Do you just have to put the cartridge in and it'll transfer all the Pokemon from that game over? Or will it only transfer over Pokemon that you've seen in that game's Pokedex (i.e. I'll have to have beaten the game and seen all the Pokemon in it)?
Because if so, then I'll just borrow my friend's Pokemon GBA games.

You need a nationaldex which you get after beating the E4. Any pokemon you have already on the GBA game can be transfered. Doesn't matter if you've seen them before. There's no way to possibly see some pokemon without first transferring.
You can also catch some version specific pokemon as well, in the wild.
Is this for Diamond only? I have pearl, I've put the honey on the tree and I spent an hour or two around the tree, without it showing up. I even tried setting my DS clock back a little just in case I missed it.

To those curious about the exclusive club in the Resort Area, don't be. All that's in there is an extremely expensive ribbon and a way to instantly max out your pokemon's happiness rating.
You need a pokemon with at least 8 ribbons to get in anyway.
So my 12 year old cousin got this and I played it today. The visualizations are cool & stuff. I played around in the Safari, where I KNEW what it was due to it being in the Red & Blue Gameboy versions.
